Background Information:

Serotonin (5-hydroxytryptamine, 5-HT) is one of the best known examples of a neurotransmitter that mediates a wide variety of physiological effects, including peripheral and central actions. At least four classes of 5-HT receptors have been distinguished pharmacologically, reflecting the second messenger system to which the receptor is coupled. The first subfamily of 5-HT1 and 5-HT5 interacts negatively with adenylyl cyclase. The second subfamily of 5-HT2 is coupled to the activation of the phospholipase C. The third subfamily, 5-HT3, is a ligand-gated ion channel. The fourth subfamily, including 5-HT4, 5-HT6, and 5-HT7, activates adenylyl cyclase to increase intracellular cAMP.

Procedure Summary:

Human recombinant serotonin 5-HT2C receptors stably expressed in CHO-K1 cells are used. Test compound and/or vehicle is preincubated with the membranes (0.036 mg/ml‡) and 1 µM GDP in modified HEPES pH 7.4 buffer for 20 minutes and SPA beads are then added for another 60 minutes at 30°C. The reaction is initiated by 0.3 nM [35S]GTPgS for an additional 45 minute incubation period. Test compound-induced increase of [35S]GTPgS binding by 50 percent or more (≥50%) relative to the 10 µM serotonin response indicates possible serotonin 5-HT2C receptor agonist activity. Test compound-induced inhibition of 0.1 µM serotonin-induced increase of [35S]GTPgS binding response by 50 percent or more (≥50%) indicates receptor antagonist activity. Note: ‡Membrane protein may change from lot to lot, the concentration used will be adjusted if necessary.
