Background Information:

b-Glucuronidase is involved in the degradation of heparin/heparan sulfate and dermatan/chondroitin sulfate. In vivo, deficiency of the enzyme is associated with the lysosomal storage disorder muco-polysaccharidosis type VII (MPS VII), or Sly syndrome, due to accumulation of partially degraded heparin/heparan sulfate and dermatan sulfate fragments. By uncoupling glucuronides, b-glucuronidase can deconjugate potential toxins, increasing the formation of carcinogens in the bowel and promoting the enterohepatic recirculation of toxins, hormones, and various drugs in the body. In fact, excessive b-glucuronidase may be a primary factor in the etiology of colon cancer.

Procedure Summary:

Bovine liver b-glucuronidase is used. Test compound and/or vehicle is preincubated with 20 μg/ml enzyme‡ in CH3COONa buffer pH 4.5 for 15 minutes at 37⁰C. The reaction is initiated by addition of 150 μM phenolphthalein glucuronide acid for another 30 minute incubation period. Determination of the amount of phenolphthalein formed is read spectrophotometrically. Compounds are screened at 10 μM.
