Background Information:

GABA is the major inhibitory transmitter in the mammallian central nervous system. GABAA receptors are anion channels gated by g-aminobutyric acid (GABA). Their amino acid sequences lie in the same superfamily as the nicotinic acetylcholine, glycine, serotonin 5-HT3 and ionic glutamate receptors, within the general class of ligand-gated ion channels. The majority of GABAA receptors carry some form of the Central Benzodiazepine subtype.

Procedure Summary:

Cerebella of Wistar derived rats weighing 175 ± 25 g are used to prepare GABAA central benzodiazepine membrane receptor in 50 mM Tris-Citrate buffer pH 7.4. A 1.5 mg‡ aliquot is incubated with 1 nM [³H]Ro-15-1788 for 120 minutes at 4℃. Non-specific binding is estimated in the presence of 10 μM diazepam. Membranes are filtered and washed, the filters are then counted to determine [³H]Ro-15-1788 specifically bound. Compounds are screened at 10 µM.
