Background Information:

Many compounds with promising pharmacological characteristics never become drugs because they are rapidly metabolized in the liver and therefore have low oral bioavailability. Clearance results obtained in vitro are also helpful in rank-ordering drug candidates based on their metabolic stabilities and assessing species and gender difference in metabolism. Eurofins has a high-throughput assay to determine metabolic stability in various matrices using LCMS analysis to quantify the percent compound remaining after incubation. Subsequently, the half-life (T1/2) is estimated from the slope of the initial linear range of the logarithmic curve of compound remaining (%) versus time, assuming first order kinetics.
