Background Information:

KRAS (Kirsten rat sarcoma virus) encodes a guanosine triphosphatase (GTPase) protein called K-Ras, acting like a switch turned on and off by the GTP and GDP molecules. Key effector pathways downstream of KRAS include mitogen-activated protein kinase (MAPK), phosphatidylinositol-3-kinase (PI3K), and Ras-like GEF (RalGEF); all these effectors are responsible for cell proliferation, cell cycle regulation, metabolic changes, cell survival, and cell differentiation. Driver mutations in KRAS underlie the pathogenesis of up to 20% of human cancers. KRASG12D variant is the most prevalent and oncogenic. Selective KRASG12D inhibitors can form salt bridge with Asp12 residue and induced-fit pocket to display anti-proliferative and anti-tumor effects.
