Background Information:
MRGPRX2 or MAS Related GPR Family Member X2 enables G protein-coupled receptor activity and neuropeptide binding activity. This receptor has been shown to be involved in calcium mobilization, mast cell degranulation and positive regulation of cytokinesis. MRGPRX2 is considered a molecular target for pharmacological intervention in mast cell-mediated allergic and inflammatory diseases. Calcium mobilization in PathHunter cell lines expressing Gq-coupled GPCRs is monitored using a calcium sensitive dye that is loaded into cells. Ligand binding stimulates GPCR activation resulting in the release of calcium from intracellular stores which leads to an increase in fluorescence in the presence of the activated calcium dye that can be measured in real time. Calcium Flux Assays are conducted in PathHunter Native cell lines expressing wild-type GPCR or in PathHunter β-Arrestin GPCR cell lines engineered to co-express the ProLink™ (PK) tagged GPCR and the Enzyme Acceptor (EA) tagged β-Arrestin.
