Background Information:

Encoded by the SCN10A gene, the Nav1.8 channel is essential for the generation and propagation of action potentials, chiefly in the dorsal root ganglion and sensory neurons called C-fibers. The channel is involved in nociception and activates at a more depolarized voltage than other sodium channels. Research has shown Nav1.8 is cold-resistant, and therefore critical to perceiving pain in cold conditions. Mutations in Nav1.8 may contribute to chronic pain conditions such as hyperalgesia, peripheral neuropathy, and allodynia. The channel is tetrodotoxin (TTX) resistant. This assay examines the effect of compound on both open-state and inactive-state channels.
