Background Information:

A colony formation or clonogenic assay has several key applications, including the assessment of irreversible cytotoxicity, reproductive toxicity, cancer/stem cell research, and therapeutic resistance.

This assay has the ability to measure cellular reproductive integrity, investigation of cancer-stemness, and is a widely used tool to predict the efficacy of particular anti-cancer therapeutics for cytotoxicity versus cytostasis. The assay will be performed in 96 well plates with 50 cells/well and treated with client compound at ten concentrations, in quadriplicates. Reference compound staurosporine or doxorubicin is used for comparison purpose in the assay. The colony is defined to consist of more than 50+ cells, which will be counted and reported as number of colonies and percent of control. Vehicle (DMSO or PBS) control will serve as percent of control values.
