Background Information:
Progesterone receptors are members of a superfamily of nuclear receptors that transduce the effects of a diverse array of small, hydrophobic ligands, including the steroid hormones, thyroid hormone, vitamin D and retinoids. In many biological systems, progestins enhance differentiation and oppose the actions of estrogens to stimulate cell proliferation. Present in two forms, PR-B and the truncated PR-A forms, neither of their physiological or pharmacological differences are known. Progesterone receptor agonists are used to suppress ovarian function (e.g., in dysmenorrhea, endometriosis, hirsutism, uterine bleeding) and to facilitate contraception and may be useful in treatment of breast cancer. Antagonists may terminate pregnancy.
PathHunter NHR Nuclear Translocation cell lines are engineered to co-express a ProLabel® (PL) tagged NHR, and Enzyme Acceptor (EA) that is localized in the cell nucleus. Activation of the receptor induces its translocation to the nucleus, forcing complementation of the two β-galactosidase enzyme fragments (EA and PL). The resulting functional enzyme hydrolyzes substrate to generate a chemiluminescent signal.
