Background Information:
Stimulation Mode: Peripheral blood mononuclear cells (cryopreserved PBMCs) are treated with client-provided reference material and the effects on T cell proliferation are determined using CellTrace™ following immuno-phenotyping into CD45+CD4+ T cells. Following initial staining with CellTrace™ PBMCs are treated with either client-provided reference material or positive proliferating inducer anti-CD3 for 96 hours. At the end of the incubation period, CD45+CD4+ T-cells are evaluated for modulation of daughter cell proliferation. Rapamycin and Upadacitinib serve has the T cell proliferation control inhibitors.

Procedure Summary:
Following treatment with client reference material, peripheral blood mononuclear cells (PBMCs) are washed and stained with fluorochrome-labeled anti-CD45, anti-CD4, and anti-CD8 antibodies. Resultant samples are analyzed on the Attune flow cytometer using a series of pre-defined analysis templates. The CD4+ and CD8+ T cell populations can be identified and quantified as relative mean fluorescence intensity (MFI) of the population of interest.
Test Sample Requirements:
Minimum sample volume for reference material treatment is 3-4 mg of dry material or 300-400 µL of 1000X highest requested testing concentration.

T cell play an essential role in the adaptive immune response. T-cells can be grouped into a variety of sub-types. The two main sub-types are the CD4+ and CD8+ T cells. CD8+ T cells are also known as "killer T cells" and they are able to directly kill virus-infected cells, as well as cancer cells. CD4+ T cells are also known as "helper T cells" and function by indirectly killing cells identified as foreign and play a key role in the secretion of cytokines that regulate the immune response.
