Antibody Characterization

Uncover High-Performing Leads from Your Candidates

Antibody Characterization

Biochemical, Functional, and Cytotoxicity Profiling

Antibody and ADC characterization uses in vitro methods to measure therapeutic antibody candidates’ binding specificity, kinetics, affinity, function, and safety. It provides essential insights into antibody properties and whether a candidate is desirable for further development. Deciding which characteristics to assess first and how to quickly funnel candidates to leads can be challenging.

Efficiently Identify High-Performing Antibody Candidates

At Eurofins Discovery, we offer a broad portfolio of biochemical, functional, cytotoxic, and other analyses that allow you to quickly and efficiently characterize your antibody and ADC candidates and prioritize those that match your target product profile. Our talented team provides a strategic characterization funnel based on your antibody concentration and needs to select promising leads from your pool of candidates. If the quantity of antibody candidates is low, our small-scale antibody expression experts can help you produce enough to complete your characterization.

Binding and Specificity Assessment

Assessing antibody-antigen binding affinity, kinetics, and specificity is essential to select the right antibody candidates for your target indication. Our team uses label-free, high-throughput surface plasmon resonance (SPR) to measure these antibody characteristics and choose when to assess binding and specificity in the characterization sequence.

Binding and Specificity Assays
  • Binding affinity measurement
  • Antigen specificity
  • Affinity and epitope binning
  • Species cross-reactivity
  • FcγR, FcRn and C1q binding affinity and kinetics

Functional Assessment

Functional characterization links antigen binding to therapeutic effects, offering early insights into mechanisms of action, efficacy, and safety. Our experienced team helps select the right functional, cell-based models and assays for your antibody or ADC’s mechanism of action and target indication.

Functional Assays
  • Cell-based binding
  • Proliferation
  • Apoptosis
  • Neutralization
  • Internalization and endocytosis

Cytotoxicity Assessment

Antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP) are cytotoxicity assays that measure the functional outcome of antibody interaction with various Fc receptors. Some candidates may use ADCC, CDC, and ADCP as therapeutic mechanisms of action, yet effector functions may be undesirable and pose a safety liability for others. Many regulatory bodies now require assessment of Fc receptor binding and functional ADCC, CDC, and ADCP assays. Our talented team can guide you through characterizing Fc binding by SPR and functional cytotoxicity assays.

Cytotoxicity Assays
  • FcγR, and FcRn and C1q binding affinity and kinetics
  • ADCC assays
  • CDC assays
