
ADCC, CDC, and ADCP Assays
Antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP) assays measure a therapeutic antibody candidate’s ability to target certain immune cells. These cytotoxicity assays determine the potential efficacy, MoA and safety of a therapeutic antibody, helping to guide antibody optimization for monoclonal antibodies (mAbs), antibody-drug conjugates (ADCs), and other antibody-based therapies.

ADCC Assays
ADCC occurs when an antibody binds to a target antigen on the surface of a target cell, distinguishing it for lysis by effector cells including natural killer (NK) cells, macrophages, or neutrophils. ADCC assays are important as they help define the MoA of an antibody candidate and can provide insight into potential efficacy or safety issues. We can develop and qualify a colorimetric, chemiluminescent, or fluorescent ADCC assays for your therapeutic antibody candidates.
CDC Assays
CDC relies on IgG or IgM antibodies binding to antigens on a target cell, triggering the complement pathway through C1q binding, formation of a membrane attack complex, and cell lysis. CDC assays assess whether a therapeutic antibody candidate can induce complement-mediated cell death, providing data on efficacy in downstream pre-clinical and clinical testing. At Eurofins Discovery, we can assess CDC activity using different methods, such as LDH release or by employing live/dead staining with viability dyes, depending on your specific requirements.
ADCP Assays
ADCP occurs when antibody-opsonized target cells activate FcγRs on the surface of macrophages, inducing phagocytosis and degradation of the target cell. ADCP assays provide data for characterizing therapeutic antibodies and understanding MoA or potential efficacy.
