Understanding the Kinetics of Your Molecule

Eurofins Discovery’s scientific experts utilize the latest in laboratory technology to perform relevant analyses for our Biophysics clients, a high-value tool for high-throughput programs. Drug discovery researchers perform kinetic analysis performed to determine the rate of complex formation and dissociation. They perform binding kinetics to calculate the speed that a molecule binds to, and dissociates from, a binding partner (antibody or receptor). Affinity (KD) describes the strength of that binding, but not the rate at which it occurs – that is described by on- and off-rate (Kon and Koff). The kinetics of a biologic is a key characteristic for the function that you want the molecule to have in vivo.
 
Full kinetic analysis is performed by Surface Plasmon Resonance (SPR) most commonly, and can be done on either one, a few, or many molecules. The target of the biologic needs to be available for the binding assay. Various tags can be used on the target protein for analysis, or the biologic can be immobilized on the biosensor and the target protein flowed over it for analysis.
