Background Information:
Melanocortins belong to the superfamily of G protein-coupled seven transmembrane proteins. G-protein-coupled receptors constitute one of the major signal transduction systems in eukaryotic cells. Coding sequences for these receptors, in those regions believed to contribute to the agonist-antagonist binding site, are strongly conserved across mammalian species. The melanocortin MC1 receptor is made primarily in melanocytes, MC2 in adrenal cortex, MC3 and MC4 almost exclusively neuronal, and MC5 in exocrine gland cells.

Procedure Summary:
This assay measures the binding of [¹²⁵I]NDP-a-MSH to human melanocortin MC4 receptors. HEK-293 cells stably transfected with a plasmid encoding the human melanocortin-MC4 receptor are used to prepare membranes in modified HEPES-KOH buffer at pH 7.0 using standard techniques. A 3.12 μg‡ aliquot is incubated with 0.02 nM [¹²⁵I]NDP-a-MSH for 120 minutes at 37°C. Non-specific binding is estimated in the presence of 3 μM NDP-a-MSH. Membranes are filtered and washed 3 times and filters are counted to determine [¹²⁵I]NDP-a-MSH specifically bound. Compounds are screened at 10 μM.
