Background Information:

Somatostatin (somatotropin-release inhibiting factor, SRIF) receptors belong to the superfamily of G protein-coupled seven transmembrane proteins. G-protein-coupled receptors constitute one of the major signal transduction systems in eukaryotic cells. Coding sequences for these receptors, in those regions believed to contribute to the agonist-antagonist binding site, are strongly conserved across mammalian species. The five known receptors can be divided into two groups: SRIF1 (SRIF1A=sst2, SRIF1B=sst5, srif1C=sst3) and SRIF2 (SRIF2A=sst1, srif2B=sst4); lower case (srif) indicates an as yet undefined physiological role. SRIF receptors are found in most peripheral tissues and within the central nervous sytem.

Procedure Summary:

This assay measures binding of [¹²⁵I]Somatostatin to human Somatostatin sst3 receptors. CHO-K1 cells stably transfected with a plasmid encoding the human Somatostatin sst3 receptor are used to prepare membranes in modified HEPES pH 7.4 buffer using standard techniques. A 0.375 μg‡ aliquot of membrane is incubated with 0.1 nM [¹²⁵I]Somatostatin for 120 minutes at 25℃. Non-specific binding is estimated in the presence of 1 µM Somatostatin. Membranes are filtered and washed 3 times and the filters are counted to determine [¹²⁵I]Somatostatin specifically bound. Compounds are screened at 10 µM.
Note: ‡Membrane protein may change from lot to lot, the concentration used will be adjusted if necessary.
