Background Information:
Bombesin receptors belong to the superfamily of G protein-coupled seven transmembrane proteins. G-protein-coupled receptors constitute one of the major signal transduction systems in eukaryotic cells. Coding sequences for these receptors, in those regions believed to contribute to the agonist-antagonist binding site, are strongly conserved across mammalian species. Bombesin receptors are widely distributed in the central nervous system and peripheral tissues. Bombesin BB2 (gastrin-releasing peptide, GRP) receptor agonism may promote gastric emptying, gastrointestinal transit, insulin secretion, inflammation, cell proliferation and tumors. Receptor antagonism may lead to abnormal social behavior.

Procedure Summary:
Human recombinant bombesin BB2 receptors expressed in human HEK-293 cells are used in modified HEPES-KOH buffer pH 7.4. A 0.1 μg‡ aliquot is incubated with 0.05 nM [¹²⁵I] (Tyr⁴)-Bombesin for 60 minutes at 25°C. Non-specific binding is estimated in the presence of 1 μM GRP (human). Membranes are filtered and washed, the filters are then counted to determine [¹²⁵I] (Tyr⁴)-Bombesin specifically bound. Compounds are screened at 10 μM.
