Background Information:
Carbonic anhydrases (CAs) are zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. At least fifteen isozymes have been described in mammals. They play key role in CO2 homeostasis, ion and acid-base balance regulation, bone resorption, calcification, renal acidification and metabolic process. Erythrocyte CAs, CA-I and CA-II, are the most well-known. The enzyme is an important target for inhibitors with applications in glaucoma, epilepsy, Parkinson’s disease, and it is being explored as a potential target for treatment of obesity and cancer.

Procedure Summary:
Human erythrocyte carbonic anhydrase I is used. Test compound and/or vehicle is preincubated with 15 µg/ml enzyme* in 50 mM MOPS solution pH 7.5 for 15 minute at 25⁰C. The reaction is initiated by addition of 500 µM 4-nitrophenyl acetate and is incubated for 30 minutes. The plate is read on the spectrophotometer (Absorbance: 400 nm). Compounds are screened at 10 µM.
