Background Information:

Cathepsin C or CTSC, also known as dipeptidyl aminopeptidase I (DPPI) or dipeptidyl transferase, is a lysosomal cysteine peptidase belonging to the papain family. Human tissues show that spleen and kidney are the richest sources of cathepsin C. In addition, serum levels are highest in hepatic diseases, followed by peripheral arterial disease, thromboembolism, myocardial infarction, diabetes mellitus, and prostatic hypertrophy. Also, cathepsin C activity is present at higher levels in cytotoxic lymphocytes and myeloid cells.

Procedure Summary:

Human cathepsin C expressed in a mouse myeloma cell line is used. Test compound and/or vehicle is preincubated with 2 μg/ml of active enzyme in modified MES buffer pH 6.0 for 15 minutes at 25°C. The reaction is initiated by addition of 20 μM Gly-Arg-AMC for another 30-minute incubation period. Determination of the amount of AMC formed is read spectrofluorimetrically at 360 nm/465 nm. Compounds are screened at 10 μM.
