Background Information:
CTSZ (Cathepsin Z, also known as cathepsin P and X or CTSX) is a cysteine protease in the papain family. It is ubiquitously expressed in human tissues and conserved in many species. Acting as a carboxypeptidase, cathepsin Z displays a unique specificity. It is ubiquitously expressed in human tissues and also distributed in cancer cells and primary tumors from different sources. Inhibition of Cathepsin Z may inhibit tumorigenesis.

Procedure Summary:
Human recombinant cathepsin Z expressed in murine myeloma NS0 cells is used. Test compound and/or vehicle is preincubated with 0.08 μg/ml enzyme‡ in modified acetate buffer pH 3.5 for 15 minutes at 25⁰C. The reaction is initiated by addition of 10 μM Mca-Arg-Pro-Pro-Gly-Phe-Ser-Ala-Phe-Lys(Dnp)-OH for another 60 minute incubation period. Determination of the amount of Mca-Arg formed is read spectrofluorimetrically at 320 nm/405 nm. Compounds are screened at 10 μM.
