In Vivo Immunization
In vivo immunization involves the introduction of a target antigen into an animal, such as a mouse or rabbit, to stimulate a natural immune response, producing antibodies. Hybridoma and Single B-Cell Recovery
Following in vivo immunization, isolating antibody-producing cells is accomplished using hybridoma techniques and single B-cell recovery. Hybridomas are generated by immortalizing antibody-producing spleen or lymph node cells, while antigen-specific single B-cells can be isolated and sequenced. Hybridomas and single B-cells can be used as a reliable source of antigen-specific antibodies for further characterization. In Vitro Display
Phage, yeast, and mammalian display techniques use synthetic or naturally-derived sequences expressed in Fab or scFv formats to enrich for antigen-specific candidates.
