Phenotypic assays evaluate a test agent by looking at its effect on specific biological endpoints, which may themselves be modulated by a number of different mechanisms or biological pathways. Examples include tumor cell viability, cytokine secretion from immune cells, and toxicities. This contrasts with target-based assays, which are also known as target engagement or mechanistic assays and focus on a specific biological target and its immediate function. Examples include kinase substrate phosphorylation and ligand-induced receptor internalization. While target-based assays inform on the specific activity of test agent at a given target, phenotypic assays inform on the broader consequences of a test agent’s action, often from a disease state perspective. However, both assay types are powerful and have complementary roles in drug discovery.
