Biological Information
Background Information:
The cysteinyl leukotrienes (LTC4, LTD4, LTE4) are products of arachidonic acid metabolism and are released from various cells, including mast cells and eosinophils. These eicosanoids bind to cysteinyl leukotriene (CysLT) receptors. CysLT receptors belong to the superfamily of G protein-coupled seven transmembrane proteins. CysLT type-1 (CysLT1) receptor is found in the human airway and on other pro-inflammatory cells.

Testing Information
Procedure Summary:
Human cysteinyl leukotriene CysLT1 receptors expressed in CHO-K1 cells are used in modified Tris-HCl buffer pH 7.4. A 5 μg‡ aliquot is incubated with 0.3 nM [³H]Leukotriene D4 for 30 minutes at 25°C. Non-specific binding is estimated in the presence of 300 nM Leukotriene D4. Membranes are filtered and washed, the filters are then counted to determine [³H]Leukotriene D4 specifically bound. Compounds are screened at 10 μM.
Note: ‡Membrane protein may change from lot to lot, the concentration used will be adjusted if necessary.
