Biological Information

Background Information:

Angiotensin receptors belong to the superfamily of G protein-coupled seven transmembrane proteins. G-protein-coupled receptors constitute one of the major signal transduction systems in eukaryotic cells. Coding sequences for these receptors, in those regions believed to contribute to the agonist-antagonist binding site, are strongly conserved across mammalian species. Angiotensin AT2 receptors are constitutively active and they are highly expressed in during fetal development. They are expressed in adult adrenal medulla, brain and reproductive tissues.

Testing Information

Procedure Summary:

Human recombinant angiotensin AT2 receptors expressed in CHO-K1 cells are used in modified Tris-HCl buffer pH 7.4. A 2.4 μg‡ aliquot is incubated with 0.05 nM [¹²⁵I] CGP-42112A for 3 hours at 37°C. Non-specific binding is estimated in the presence of 10 μM (Sar¹, Ile⁸)-Angiotensin II. Membranes are filtered and washed, the filters are then counted to determine [¹²⁵I] CGP-42112A specifically bound. Compounds are screened at 10 μM.
Note: ‡Membrane protein may change from lot to lot, the concentration used will be adjusted if necessary.

Clinical Relevance

Therapeutic Area:

CNS/Addiction/Pain

Adverse / Beneficial Action:

AT2 receptors are upregulated after vascular injury, cardiac failure and wound healing. AT2 receptor agonism may result in hypotension while antagonism may facilitate cell proliferation, inhibit and inhibit apoptosis.
